ABSTRACT: The main method of sugarcane smut (Sporisoriumscitamineum)control is the genetic resistance. This study assessed the reaction of Brazilian sugarcane genotypes to the smut. The design used was randomized blocks, with 41 genotypes and four replications. The experimental unit consisted of five seedlings per genotype. The genotypes were inoculated by immersing the buds in a smut spores’ suspension. For each genotype, it was obtained the disease incidence in percentage of stalk with whip and with the disease incidence data over time the area under the disease progress curve (AUDPC) were calculated for each genotype. Based in the AUDPC, three genotypes were classified as highly resistant (7.3%), eighteen as resistant (43.9%), twelve as moderately resistant (29.3%), three as moderately susceptible (7.3%) and five as susceptible (12.2%). Genotypes with higher levels of resistance to smut sugarcane can be used for integrated disease management and can be used as parents in new crosses in order to obtain progenies with higher proportion of genotypes resistant to smut.

RESUMO: O principal método de controle do carvão da cana-de-açúcar (Sporisorium scitamineum)é o controle genético. O objetivo deste estudo foi avaliar a reação de genótipos brasileiros de cana-de-açúcar ao carvão. O experimento foi conduzido em blocos casualizados, com 41 genótipos e quatro repetições. A unidade experimental foi composta por cinco mudas por genótipo. Os genótipos foram inoculados através da imersão das gemas em uma suspensão de esporos do fungo. Para cada genótipo, obteve-se a incidência da doença em percentagem de colmos com chicote de carvão e com os dados de incidência da doença ao longo do tempo foi calculada a área abaixo da curva de progresso da doença (AACPD) para cada genótipo. Baseado na AACPD, três genótipos foram classificados como altamente resistentes (7,3%), dezoito como resistentes (43,9%), doze como moderadamente resistentes (29,3%), três como moderadamente suscetíveis (7,3%) e cinco como suscetíveis (12,2%). Genótipos com os maiores níveis de resistência ao carvão da cana-de-açúcar que podem ser utilizados para o manejo integrado da doença e como genitores em novos cruzamentos para obter progênies com maior proporção de genótipos resistentes ao carvão.

RESUMO: A análise de regressão logística é uma técnica que pode auxiliar programas de melhoramento genético na seleção de clones, especialmente nos estágios iniciais, em que a precisão experimental é baixa. Este trabalho teve como objetivo identificar os principais caracteres agronômicos para a seleção clonal de cana energia e verificar a eficiência do modelo logístico na predição dos genótipos a serem selecionados. As avaliações foram realizadas em 220 clones na primeira soca. Os dados foram submetidos à análise de regressão logística binária. O número de colmos por metro foi um caractere de maior importância na seleção de clones de cana energia. Além disso, os clones com nota menor para incidência de carvão tiveram maior chance de serem selecionados. As capacidades preditivas dos modelos qualitativos e quantitativos foram de 94% e 88%, respectivamente. O uso de um modelo qualitativo mostrou-se eficaz na previsão do número de genótipos de cana energia a serem selecionados e pode ser usado como estratégia de seleção.

ABSTRACT: Logistic regression analysis is a technique that may aid genetic breeding programs in the selection of clones, especially in the early stages where experimental accuracy is low. This research aimed to identify the most important agronomic traits for energy cane clonal selection, and to verify the efficiency of the logistic model in predicting the genotypes to be selected. Evaluations were carried out on 220 clones in the first ratoon. The data were subjected to binary logistic regression analysis. Stalk number per meter was the most important trait in the selection of energy cane clones. In addition, plants with lower grade for smut incidence had a greater chance of being selected. The predictive capacities of the qualitative and quantitative models were 94% and 88%, respectively. The use of a qualitative model proved to be effective at predicting the number of energy cane genotypes to be selected and could be used as a selection strategy.

Abstract Cultivar RB036091 is an early-maturing sugarcane with a long period of industrial suitability; in the Central South region of Brazil, harvest is recommended between April and August, and the cultivar is indicated for planting on moderate to highly fertile soils. It is widely adaptable and has a high sugar yield and stability of agricultural yield.

RESUMO: O objetivo desse trabalho foi selecionar famílias e clones de cana-de-açúcar tolerantes ao estresse por frio. As avaliações ocorreram durante três fases de seleção no município de Pelotas, no estado do Rio Grande do Sul, Brasil. Os experimentos foram conduzidos no delineamento de blocos incompletos, sendo inicialmente composto por 4.452 seedlings oriundos de 53 famílias de irmãos completos. Além dos caracteres teor de sólidos solúveis (BRIX), tonelada de colmos por hectare (TCH) e tonelada de brix por hectare (TBH), foi avaliada a sobrevivência do meristema apical (GEM) na primeira fase de seleção (T1) do programa de melhoramento genético. Ao final de três fases de seleção, 15 clones pertencentes a 14 das 53 famílias avaliadas na primeira fase (T1) foram selecionados para a fase de experimentação, destes, destacam-se: RS/PR126066, RS/PR126044, RS/PR126052, RS/PR126007 e RS/PR126033, pois além de apresentar desempenho satisfatório para os caracteres TCH e TBH, foram avaliados com notas máximas (N4 e N5) para sobrevivência de gema apical no primeiro ciclo de seleção.

ABSTRACT: The purpose of this study was to select cold-tolerant sugarcane families and clones. Evaluations were carried out during three selection phases in the municipality of Pelotas, state of Rio Grande do Sul, Brazil. The experiments were arranged in an incomplete block design, with initially 4,452 seedlings of 53 full-sib families. Aside from the traits soluble solids content (BRX), tons of stalks per hectare (TSH) and tons of brixper hectare (TBH), the survival of the apical bud (ABS) was evaluated in the first selection stage (T1) of the breeding program. At the end of three selection phases, 15 clones of 14 of the 53 families evaluated in the first phase (T1) were selected for the experimental phase. Of these, the clones RS/PR126066, RS/PR126044, RS/PR126052, RS/PR126007 and RS/PR126033, had a good performance for apical bud survival in the first selection phase.

Abstract The sugarcane cultivar RB036066 is medium-maturing and has a long period of industrial suitability; in the South-Central region, harvest is recommended between June and September, and it is indicated for planting on medium to highly fertile soils. The cultivar is widely adaptable and has high sugar yield and stability of agricultural production.

RESUMO Esse trabalho teve como objetivo avaliar o efeito de vários fatores que podem afetar a transformação genética de explantes cotiledonares de Eucalyptus saligna mediante co-cultura com a cepa EHA105 de Agrobacterium tumefaciens. Foi utilizado o vector pBI121 carregando o gene repórter gus sob controle do promotor 35SCaMV. Foi estudado o efeito dos fatores a seguir: pré-cultura dos explantes, uso de diferentes antibióticos e presença de acetosiringona (AS) nos meios de co-cultura. Uma solução contendo ácido ascórbico (250mg.L-1), ácido cítrico (25mg.L-1) e PVP-40 (1g.L-1) foi utilizada no momento da excisão dos explantes. A pré-cultura dos explantes antes da co-cultura com a bactéria foi de 4 dias, em meio de cultura MS suplementado de 4,4µM BAP e 2,7ìM ANA. Após a co-cultura três concentrações de canamicina (12,5; 25 e 50mg.L-1) combinadas com 300mg.L-1 de Augmentin® no meio de cultura foram testadas. O efeito do Augmentin® (300mg.L-1) nos explantes foi comparado ao da cefotaxima (500mg.L-1), em meio contendo 50mg.L-1de Km. Foi concluído que Augmentin® estimula a organogênese, em comparação com a cefotaxima, que a concentração de 12,5mg.L-1 de Km permite a seleção de explantes transformados com o gene gus e, finalmente, a adição de AS (50ìM) aos meios de co-cultura líquido e sólido tem efeito positivo sobre a expressão do gene gus. O uso de antioxidante não é necessário e a pré-cultura dos explantes antes da inoculação com A. tumefaciens não tem efeito na regeneração de gemas nem na expressão do gene gus. A eficiência de transformação foi de 1,5%.

ABSTRACT This study aimed to evaluate the effect of factors that may affect the genetic transformation of cotiledonary explants of Eucalyptus saligna mediated by EHA105 strain of Agrobacterium tumefaciens. The vector pBI121 carrying gus gene under control of 35S CaMV promoter was used. The effect of the following factors was evaluated: explant pre-culture, use of different antibiotics and presence of acetosyringone (AS) in co-culture media. An antioxidant solution was also used during excision, containing ascorbic acid (250mg.L-1), citric acid (25mg.L-1) and PVP-40 (1g.L-1). Pre-culture of the explants before the co-culture with bacteria was done over a 4-day period in MS culture medium supplemented with 4.4µM BAP and 2.7ìM NAA. After theco-culture period, three concentrations of kanamycin (12.5;25 and 50mg.L-1) combined with 300mg.L-1 Augmentin® in the culture medium were tested The influence of the antibiotic was also evaluated by keeping the explants in a medium containing 50mg.L-1 Km and 300mg.L-1 Augmentin® or 500mg.L-1 cefotaxime. It was concluded that Augmentin® stimulates organogenesis, that a Km concentration of 12.5mg.L-1 allows selection of explants transformed with gus gene and, finally, the addition of AS (50ìM) to the liquid and solid co-culture media has a positive effect on gus gene expression. Moreover, the use of an antioxidant solution during cotyledon excision is dispensable and the pre-culture of the explants has no effect on bud regeneration or gus gene expression. A transformation efficiency of 1.5% was reached.

Para a avaliação de experimentos com cana-de-açúcar, tem-se recomendado a colheita total de parcelas, algo que, devido aos seus custos, tem se mostrado inexequível. Alternativamente, pode-se estimar a produção com base na tomada de amostras que representem as parcelas. O objetivo deste trabalho foi estudar a suficiência amostral em cana-de-açúcar para a estimação da massa dos colmos de parcelas experimentais. Reamostragens com reposição subsidiaram o uso dos modelos da curva potencial, linear e quadrático segmentados com resposta platô e do modelo do coeficiente de variação relativo. Também foram realizados estudos simulados de análise de variância. Os resultados indicaram que amostras de 20 colmos coletados aleatoriamente na parcela experimental permitem estimar adequadamente a variável massa do colmo. Com este valor, serão obtidas avaliações confiáveis da produtividade em colmos

During the evaluation of the experimental phase, which corresponds to competition experiments and indication of cultivars, is recommended total harvest of plots, which, owing to their costs, can become impracticable. Optionally, one can estimate the production based on taking samples representing plots. The aim of this study was to research the sample sufficiency in sugarcane for estimating the mass of stalks of experimental plots. One used the maximum curvature of the power model, the linear and quadratic segmented models with response plateau and the relative CV model. Studies based on simulated analysis of variance were done also. The application of these models and the ANOVA was supported by bootstrap resamplings. The results indicated that a sample made by 20 stalks harvested in plot can be used for estimating properly the variable mass of the stalk. This value may be used for estimating the sugarcane yield per hectare

RESUMO Os objetivos deste trabalho foram avaliar diferentes meios de cultura na organogênese indireta e na multiplicação in vitro de brotos de Eucalyptus benthamii x Eucalyptus dunnii. Para organogênese, explantes foliares foram excisados no sentido transversal e cultivados in vitro, sendo os seguintes fatores testados: dois meios de cultura (MS N/2 e JADS) adicionados de 0,1 µM de ANA, duas concentrações de thidiazuron (0,1 e 0,5 μM) e presença ou não de PVP-40 (250 mg L-1). Após 70 dias de cultivo foram avaliadas as porcentagens de explantes oxidados totalmente, formando calo, produzindo antocianina, formando gema, formando brotações e o número de brotações formadas por explante regenerando. No experimento de multiplicação, brotações isoladas foram cultivadas em meio MS, JADS e WPM, adicionados de 1,11 μM de BAP. Foram realizados quatro subcultivos a cada 28 dias e em cada subcultivo foram avaliados: a porcentagem de oxidação, de explantes apresentando clorose total ou parcial, massa fresca e número médio de brotos por explante. O meio de cultura MS N/2 suplementado com 0,1 µM de ANA, 0,5 µM de TDZ e PVP-40 promoveu a maior taxa de organogênese (8,3%). No meio de cultura MS com 1,11 μM de BAP, a taxa de multiplicação foi maior que nos outros meios, no primeiro e segundo subcultivos (9,28 e 9,24 por mês), não havendo diferença entre os três meios nos demais subcultivos.

ABSTRACT The aims of this research were to evaluate different culture media for indirect organogenesis and shoot multiplication of Eucalyptus benthamii x Eucalyptus dunnii. For organogenesis, leaf explants were used to test the following treatments: two culture media (MS N/2 and JADS) supplemented with 0.1 µM 1-naphthaleneacetic acid (NAA) and thidiazuron (TDZ) (0.1 or 0.5 μM), with or without PVP- 40 (250 mg L-1). The percentage of oxidized explants, callus forming explants, explants with anthocyanin, buds, shoots and the shoot number per explant were evaluated. In the multiplication experiment, isolated shoots were cultivated in MS, JADS and WPM media, all supplemented with 1.11 μM BAP. Four subcultures were carried out every 28 days. In every subculture the explant oxidation, partial or total leaf chlorosis, fresh mass and mean number of shoot per explant were evaluated. The MS N/2 medium supplemented with 0.1 µM NAA and 0.5 µM TDZ promoted the highest rate of organogenesis (8.3%) and the culture media MS supplemented with 1.11 μM BAP the multiplication rate was higher than in the other media, in the first and the second subcultures (9.28 and 9.24, respectively), without differences between the three media in the following subcultures.

O desenvolvimento de um protocolo eficiente em um sistema de regeneração é crucial para a transformação genética de cana-de-açúcar. Os objetivos deste estudo foram a regeneração de plantas a partir de embriões somáticos e a análise da origem e modelos de divisão celular durante os diferentes estágios dos embriões somáticos, das cultivares de cana-de-açúcar RB855156 e RB72454. Em ambas as cultivares, os melhores resultados para a indução de calos embriogênicos foram obtidos com o cultivo dos explantes em meio de cultura contendo 13,5 µM de 2,4- ácido-diclorofenoxiacético (2,4-D), seguido do cultivo dos calos em meio de cultura sólido com 4,5 µM de 2,4-D. As maiores taxas de indução de brotações foram encontradas com 8,9 de 17,8 µM benzilaminopurina (BAP) para RB855156 e 17,8 µM BAP para RB72454. O meio de Murashige and Skoog (1962) (MS) com 2,5 µM de ácido indolbutírico (IBA) para RB855156 e 5,0 µM IBA para RB72454 foram adequados para o enraizamento das brotações. As mudas foram aclimatizadas com sucesso e a taxa de sobrevivência foi de 100% para ambas as cultivares. As análises histológicas revelaram que a origem das brotações em ambas as cultivares ocorreu via embriogênese somática.

The development of an efficient regeneration system is crucial for the genetic transformation of sugarcane. The aims of this study were to regenerate plants from somatic embryos and analyze the origin and division pattern of cells during the different embryonic developmental stages of the sugarcane cultivars RB855156 and RB72454. For both cultivars, the best results for embryogenic callus induction were obtained with explant cultures on culture medium containing 13.5 µM 2,4-dichlorophenoxyacetic acid (2,4-D) followed by callus culture on solid medium with 4.5 µM 2,4-D. A higher rate of shoot induction was observed in RB855156 with the addition of 8.9 and 17.8 µM benzylaminopurine (BAP); this higher rate was also observed in RB72454 with the addition of 17.8 µM BAP. Murashige and Skoog (1962) (MS) medium containing 2.5 and 5.0 µM indolebutyric acid (IBA) for RB855156 and RB72454, respectively, was suitable for rooting. The plantlets were successfully acclimatized and the plant survival rate was 100% for both cultivars. Histological analysis revealed that the shoots in both cultivars originated from somatic embryogenesis.

O objetivo deste trabalho foi avaliar genótipos de cana-de-açúcar, considerando toneladas de pol por hectare (TPH), estratificando cinco ambientes de produção no Paraná. Foram analisados 20 genótipos e dois padrões, em três safras consecutivas. Os métodos estatísticos utilizados foram AMMI e GGE Biplot. O GGE Biplot agrupou os locais em dois mega-ambientes e apresentou quais genótipos estiveram entre os melhores para cada mega-ambiente, facilitando a seleção dos genótipos superiores. Outra vantagem do GGE Biplot foi a representação do genótipo e do ambiente ideal, que serviram de referência para a avaliação dos genótipos e para escolha de ambientes com maior interação GxE. Ambos os modelos, mostraram que os genótipos mais produtivos em TPH, foram: RB006970, RB855156 e RB855453 e que o ambiente São Pedro do Ivaí apresentou maior interação GxE. Ambas metodologias apresentaram elevada porcentagem de explicação das soma dos quadrados, tendo a metodologia AMMI uma pequena vantagem sobre o GGE Biplot.

The purpose of this study was to evaluate sugarcane genotypes for the trait tons of sugar per hectare (TSH), stratifying five production environments in the state of Paraná. The performance of 20 genotypes and 2 standard cultivars was analyzed in three consecutive growing seasons by the statistical methods AMMI and GGE Biplot. The GGE Biplot grouped the locations into two mega-environments and indicated the best-performing genotypes for each one, facilitating the selection of superior genotypes. Another advantage of GGEBiplot is the definition of an ideal genotype (G) and environment (E), serving as reference for the evaluation of genotypes and choice of environments with greater GE interaction. Both models indicated RB006970, RB855156 and RB855453 as the genotypes with highest TSH and São Pedro do Ivai as the environment with the greatest GE interaction. Both approaches explained a high percentage of the sum of squares, with a slight advantage of AMMI over GGE Biplot analysis.

O objetivo deste trabalho foi testar métodos para avaliação da germinação na pré-colheita de cultivares de trigo. Quatorze cultivares de trigo foram cultivadas em Londrina e Ponta Grossa, PR. Aos 10 e 17 dias após a maturidade fisiológica, elas foram colhidas e avaliadas pelos métodos de germinação por simulação de chuva (em casa de vegetação), com grãos nas espigas e com grãos removidos das espigas. O método do grão na espiga permitiu a diferenciação das cultivares, porém mostrou graus de resistência distintos das descrições disponíveis para as cultivares. O procedimento do grão removido da espiga não possibilitou a distinção segura dos dados obtidos quanto à germinação, em qualquer época ou local de coleta. O método por simulação de chuva é o mais adequado para avaliação de cultivares quanto à germinação na pré-colheita, independentemente da época de coleta e do local avaliado.

The objective of this work was to test methods for pre-harvest sprouting assessment in wheat cultivars. Fourteen wheat cultivars were grown in Londrina and Ponta Grossa municipalities, Paraná state, Brazil. They were sampled at 10 and 17 days after physiological maturity and evaluated using the methods of germination by rainfall simulation (in a greenhouse), in-ear grain sprouting, and grains removed from the ears. The in-ear grain sprouting method allowed the differentiation of cultivars, but showed different resistance levels from the available description of cultivars. The sprouting of grain removed from the ears did not allow a reliable distinction of data on germination in any harvest date or location. The method of rainfall simulation is the most suitable for the assessment of cultivars as to pre-harvest sprouting, regardless of the sampling date and evaluated location.

O híbrido Eucalyptus grandis x Eucalyptus urophylla apresenta alta produtividade e potencial para indústrias de papel, celulose e fibras. A organogênese de explantes foliares de dois clones de E. grandis × E. urophylla foi estudada para verificar fatores como tempo de repicagem das plântulas matrizes em meio de multiplicação; tipo de explantes, folhas inteiras e de meias-folhas (porções basais e apicais) e dos dias que os explantes foliares permaneceram em meio de regeneração. Foram observadas diferenças na capacidade organogênica dos dois clones testados. A parte basal das folhas, coletadas de brotações repicadas a cada 17 dias, apresentou superioridade organogênica. Explantes foliares devem ser transferidos para novo meio de indução de brotações a cada cinco dias. Este estudo também teve como objetivo avaliar fatores que afetam a transformação genética de explantes foliares com o gene uidA, via co-cultivo com Agrobacterium tumefaciens, tais como a pré-cultura, a duração da co-cultura e a adição da acetoseringona no meio de cultura. As melhores condições para a expressão do gene uidA foram a cada dois dias de pré-cultura de explantes foliares, três dias de co-cultura com a bactéria e a adição de acetoseringona nos meios de pré e co-cultura.

The hybrid Eucalyptus grandis x Eucalyptus urophylla presents high levels of productivity and potential for use in paper, cellulose and fiber industries. The bud organogenesis from leaf explants of two clones of E. grandis × E. urophylla was studied in order to verify the effect of several factors: subculture duration on multiplication medium, type of explants, entire and half leaves: basal and apical portions, and duration of the culture on a regeneration medium. Differences in organogenic capacity of the two clones tested were observed. The explant most recommended for organogenesis is the basal section of the leaf collected from shoot clusters subcultured every 17 days. Moreover, the leaf explants must be transferred to a fresh bud induction medium every five days. This study also aimed at evaluating factors affecting the genetic transformation of leaf explants with the uidA gene, via co-cultivation with Agrobacterium tumefaciens, such as the pre-culture of the explants on a specific medium, the duration of their co-culture with the bacteria and the addition of acetosyringone to the culture media. The best conditions for the expression of the uidA gene were two days of pre-culture of the leaf tissues, three days of co-culture with the bacteria and the addition of acetosyringone in pre- and co-culture media.

O objetivo deste trabalho foi comparar a seleção via procedimento BLUP individual simulado (BLUPIS) versus seleção massal em famílias de irmãos-completos de cana-de-açúcar. Foram utilizadas 80 famílias originadas de cruzamentos biparentais da série RB03. O experimento foi desenvolvido em área experimental, localizado no Município de São Tomé, Paraná. O delineamento experimental foi em blocos incompletos, com cinco repetições por família. Para seleção considerou-se a variável tonelada de sólidos solúveis por hectare (TSH). Utilizou-se o modelo 35 do programa computacional Selegen REML/BLUP, via procedimento BLUPIS para a indicação do número de clones potenciais a serem selecionados. Estes resultados foram correlacionados com o número de clones selecionados via seleção massal dentro das famílias. A seleção clonal via procedimento BLUPIS indica maior número de clones promissores para caracteres quantitativos dentro de famílias com elevados efeitos genotípicos.

The aim of this work was to compare the selection by simulated individual BLUP procedure (BLUPIS) versus individual selection in full-sib sugarcane families. Eighty families originated from bi crosses of RB03 series were used. The experiment was carried out in an experimental area in São Tomé County, Paraná State. The experimental design was in incomplete blocks, with five replications per family. For selection, the variable soluble solids per hectare (TSH) was used. For indication of the number of potential clones to be selected, the model 35 of the software Selegen REML/BLUP by BLUPIS procedure was used. The results of BLUPIS were correlated with the number of selected clones by individual selection. The results showed that clone selection by BLUPIS procedure can indicate higher number of promising clones for quantitative trait inside families with high genotypic effects.

RB966928 is an early to medium maturing sugarcane cultivar and in Central-Southern Brazil it is harvested between April and June; it is recommended for planting in environments with medium to high soil fertility. It stands out with a high ratoon yield and with excellent sprouting in both plant cane and ratoon cane. It is tolerant to the major diseases of economic importance of sugarcane.
